چکیده

Pure strains of Purple Non-Sulfur (PNS) bacteria: Rhodobacter sphaeroides strain O.U 001 was studied for its efficiency to produce biohydrogen using the photofermentative pathways. An annular photobioreactor was being designed aiming the uniform light distribution. In prototype annular photobioreactor, at an initial pH of 6.8±0.2, temperature 32 ± 2 ◦C, inoculum volume 10% (v/v), inoculum age of 48 h, light intensity of 15±1.1 W m and stirring at 250 rpm; the average H2 production rate was about 6.55±0.05 ml H2 h l media for Rhodobacter sphaeroides strain O.U 001 (initial Malic acid concentration of 2.01 g l). The biohydrogen production was found to be microbial mixed growth associated function. Keywords-Biohydrogen, photobioreactor, photofermentation, purple non-sulfur (PNS) bacteria.

Hydrogen generation by Rhodobacter spahaeroides O.U. 001: the effect of photobioreactor construction material

Genertation of hydrogen by purple, non-sulfur bacteria of Rhodobacter sphaeroides O.U.001 was tested in photobioreactors made of different materials. Reactors made of ordinary sodium-type glass, borosilicate glass (Pyrex) or polycarbonate were applied in the present study. The applied medium containing malic acid and sodium glutamate inoculated with 0.11 or 0.31 g dry wt. of Rhodobacater sphaer...

Heterologous expression of glutamyl-tRNA reductase gene in Rhodobacter sphaeroides O.U.001 to enhance 5-aminolevulinic acid production

The pathways for synthesis of 5-aminolevulinic acid (5-ALA) use either succinyl-CoA and glycine (C-4 pathway), or glutamate (C-5 pathway). Although Rhodobacter sphaeroides synthesizes 5-ALA through the C-4 pathway, it also has the genes coding for the enzymes of the C-5 pathway, except for glutamyl-tRNA reductase. The glutamyl-tRNA reductase gene was cloned from Rhodospirillum rubrum and expres...

Novel properties of photofermentative biohydrogen production by purple bacteria Rhodobacter sphaeroides: effects of protonophores and inhibitors of responsible enzymes

BACKGROUND Biohydrogen (H2) production by purple bacteria during photofermentation is a very promising way among biological H2 production methods. The effects of protonophores, carbonyl cyanide m-chlorophenylhydrazone (CCCP), 2,4-dinitrophenol (DNP), and inhibitors of enzymes, involved in H2 metabolism, metronidazole (Met), diphenyleneiodonium (DPI), and dimethylsulphoxide (DMSO) on H2 producti...

Performance, 5-aminolevulinic acid (ALA) yield and microbial population dynamics in a photobioreactor system treating soybean wastewater: Effect of hydraulic retention time (HRT) and organic loading rate (OLR).

Effects of hydraulic retention time (HRT) and influent organic loading rate (OLR) were investigated in a photobioreactor containing PNSB (Rhodobacter sphaeroides)-chemoheterotrophic bacteria to treat soybean wastewater. Pollutants removal, biomass production and ALA yield in different phases were investigated in together with functional microbial population dynamics. The results showed that pro...

Optimization of Growth of Rhodobacter Sphaeroides Using Mixed Volatile Fatty Acidsby Response Surface Methodology

A combination of photosynthetic bacteria along with anaerobic acidogenic bacteria is an ideal option for efficient hydrogen production. In the present study, the optimum concentration of substrates for the growth of Rhodobacter sphaeroides was found by response surface methodology. The optimum combination of three individual fatty acids was determined by Box Behnken design. Increase of volatile...
